Coombs tests: direct vs. indirect
Coombs tests are classified into two types: direct Coombs testing and indirect Coombs testing.
- The direct antiglobulin test (DAT) is more often used and looks for antibodies on the surface of your red blood cells. The Coombs reagent causes antibody-bound red blood cells to cluster together.
- The indirect antiglobulin test (IAT) looks for antibodies floating in the serum, which is the liquid portion of your blood. To do this, your red blood cells are removed from the sample and replaced with foreign red blood cells that attract antibodies. If the reagent causes these new cells to cluster together, it indicates that antibodies are present.
